The Turtles are fighting in the lair deep beneath New York City, Raphael versus Donatello and Leonardo versus Michelangelo. After winning their respective bouts, Leonardo and Raphael engage in a duel, with Raphael emerging victorious. Splinter congratulates them all on their excellent performance and gives Raphael a nerve pinch to make him feel less arrogant for boasting about his triumphs. Splinter is a little amused when Raph swiftly and passionately remarks on how well his brothers performed.

Following Michelangelo's entreaties, Splinter recounts how the Turtles became a family as they celebrate their Mutation Day over a dinner of worms and algae. He purchased four newborn turtles from a pet shop one day when he was still a human. Sensing something "off" about the stranger, he chose to follow him after bumping into him on the street. He discovered the guy trading a mutagen canister with another odd man. A mouse suddenly screamed, revealing his whereabouts. A brawl broke out as the weird individuals attacked, shattering the canister in the process. They preserved the broken container ever since Splinter and the four young turtles transformed.

Leonardo watches his favorite show, Space-Heroes, before leaving the house, imitating the words and gestures of the characters. Leonardo aspires to be a great hero like Captain Ryan from the show, but Raphael mocks him. They then get ready to depart after Donatello and Michelangelo join them. Splinter sends them off with a lot of advice, but he specifically tells them to avoid humans. The Turtles are captivated by the metropolis once they emerge on the surface. They soon come across a pizza delivery boy.

A pizza box slips off his bike and hits the ground when Raphael scares him away. Curious, the gang takes the pizza with them when they move from the street to the roofs where they are less visible. Michelangelo offers to sample the odd dish first. In addition to being safe, it tastes great, and the turtles quickly gobble it up.

Leonardo believes it's time to head home after more roof hopping, but Donatello is instantly enamored with a teenage girl he sees on the street with her father. A van suddenly pulls up in front of the two people, and other odd individuals start to walk toward them. Although Leonardo is hesitant to defy Splinter's commands to avoid people, he is compelled to do so once the other three do. The Turtles save the girl just before she and her father are dragged into the van, but they continue to obstruct each other during the altercation. The girl's fear of them is equal to that of her assailants.
Michelangelo unintentionally strikes Donatello in the head with his nunchucks just as he is beginning to win her trust. The van then leaves after they are both knocked down and the men kidnap the girl. Donatello, Leonardo, and Raphael follow them.
As Michelangelo begins to follow them, one of the villains confronts him. He tries to flee after his nunchucks fail, but he is stuck in an alley. He is startled to see that his assailant is a robot with an alien brain in his torso as he swings his kusarigama wildly at him. Michelangelo catches up with his brothers after a brief battle with the extraterrestrial, but they don't believe him when he tells them what he saw.

After Splinter chastises them for their failure and they all begin blaming one another, the Turtles retreat home in humiliation. When Mikey unintentionally places the blame on Splinter, he corrects him, saying that as their father and instructor, he should be held accountable because Splinter trained them as individuals rather than as a team. After telling them that they are still not prepared for the surface world, he thinks about giving them another year of training before making another attempt. The abducted victims can't wait that long, Donnie protests. The sensei is moved by his son's remarks and looks at a picture of himself, a woman, and their newborn child that is on the shelf behind him. He can still hear the fire.

Leo has the crew stake out a structure with the same emblem as the kidnappers' van back on the surface. He thinks that if they wait long enough, the bad guys will eventually come to question them. After two hours, the same van from earlier arrives just as Raph is beginning to gripe that they are wasting their time. Leo starts to deliver Captain Ryan's monologue about his "bold and daring" plan, but he has to play catch-up because his team has already set off. The Turtles swarm the driver as soon as he exits the vehicle, but he catches them off guard with a portable laser rifle and quickly drives away in the van. Then the Turtles fly to the roofs.

Raphael attempts to jump onto the van's top, but a sudden swerve sends him smashing into a wall. The driver shoots his odd weapon out the window at the remaining Turtles, keeping them at away. The vehicle eventually crashes and topples over after Leonardo uses a well-aimed throwing star to burst one of its tires. He utilizes hand motions to tell his siblings to come from the back when the Turtles are able to go much closer to investigate; this would be a more effective maneuver if his brothers understood what he was saying. The van's back doors open as they approach, and a glowing canister suddenly slides out and stops.
Raphael attempts to jump onto the van's top, but a sudden swerve sends him smashing into a wall. The driver shoots his odd weapon out the window at the remaining Turtles, keeping them at away. It finally crashes and topples over as Leonardo throws a throwing star. He signals his brethren to come from the back with hand gestures once the Turtles are close enough to investigate. The van's rear doors open as they approach, and a luminous canister suddenly emerges and comes to a halt at Michelangelo's feet. Mikey exclaims in shock, "MOM?!?" after realizing it resembles the drug that changed them fifteen years ago.

This is a great way to kick off TMNT. The episode really gets deep into the back story and shows clearly how the turtles mutated from turtles to human turtles. Also, splinter's back story is shown well-being a former ninja sensei. The turtles are introduced well too with the action that they get into, showing how each one is unique, special, and had abilities that make the team stronger as each turtle is different, but splinter urges them to put their unique abilities together to work as a team.
